Teacher can be " lo si" or "seen san". Is there a difference? Is there a more polite way to call someone? I used to be taught to say ___ seen san, but I don't know if it only applies to middle school teachers, etc.

Lou si is more affectionate nowadays than seen saang, it seems to me.
Seen saang is also used to refer to adult males whose name you don't know. Kinda like in English, some people address a male stranger as "Sir" or less politely "Mister".
